Overview

Development of novel ZFP-based gene therapy approaches for monogenic disorders.

The Wolfe laboratory will be filling a postdoctoral position to develop new gene therapy reagents for the treatment of monogenic disorders in collaboration with other laboratories within UMMS. These efforts will focus on the creation of novel Cys2His2 zinc finger protein-based therapeutics for delivery to affected cell types. This project will focus on:

1) The development of Cys2His2 zinc finger proteins (ZFPs) with novel DNA binding specificity to target specific (therapeutic) genomic loci.

2) The application of these ZFPs in cell culture and in vivo models of disease to abrogate disease-associated phenotypes.

These studies will expose the successful candidate to cutting-edge ZFP design and selection approaches, as well as in vitro and in vivo delivery approachs. The successful candidate will also learn cutting edge deep-sequencing approaches to monitor the genome-wide binding locations and impact of the ZFPs on the genome and transcriptome. The successful candidate will have the opportunity to participate in meetings/conferences associated with gene therapy, which will broaden their exposure to cutting edge research within the field. In addition, the successful candidate will learn to utilize CRISPR-Cas9 and CRISPR-Cas12a nuclease technologies that are being developed in the Wolfe laboratory. Candidates should have a Ph.D. in biology, biochemistry, chemistry or a related biomedical science. In addition, the potential candidate must have strong oral and written communication skills and be eager to present research results at national meetings. Prior experience with Cys2His2 zinc finger proteins, cell culture, and deep-sequencing technologies is advantageous, but not required.
